What can I expect to pay to fly to Quenca from Quito? What is the airline called. Is it worth the busride? Are there comionetas(vans) travelling between cities like you see in Guatemala?

I took a plane one-way (on AeroGal) and paid about $65 last month. The bus ride is less than $10 but takes 8-10 hours. I found the average bus in Ecuador to be much nicer than the average bus in Guatemala (no old US school buses) but they are far from luxurious. It's really a question of time vs money. The plane ride takes less than 45 minutes.
